Strains that constitutively express ß-galactosidase

There is another type of mutation that we are interested in, namely those that lead to a loss of normal regulation of ß-galactosidase expression.

Mutations of this class would lead to cells making ß-galactosidase constitutively, i.e. in the absence of lactose or other inducers.

How can we identify such mutations?



What does it mean if there are white colonies when you plate out a rob- strain on an X-GAL plate?
